The answer could be in the question? If indeed being in pursuit of pleasure is the essence of human nature, then the question is the answer. However, Buddha believed that to reach "enlightenment", the individual must avoid pleasure: "the cause of all suffering is desire, to end suffering, is to end desire". Of course this doesnt mean that seeking pleasure isn't intrinsic, but it does suggest that to reach ultimate pleasure, we must avoid seeking it. Ironic?

I'm not so sure that all humans are pleasure seekers. But the answer usually given by Christianity that is sin plays a big part in men treating pleasure as the Good. Of course, pleasure in itself is not evil or pernicious. But hedonism is another story.

We still have rewards in the release of endorphins in the brain for doing certain things. It was natures way of making sure we didnt become extinct in our cave man days but we still follow these very old patterns even today. Eating is one and sex to. Certain drugs play around with this area of the brain to like cocaine.
Other things like football or anything physical or mentally exciting releases adrenalin which also makes us feel great.
We used back then to give us extra energy when fighting something with more teeth than us. Theres nothing spiritual to it. Its all biology!

Because pleasure is the universe's way of telling us we are doing 'right'. If it hurts it is obviously ‘wrong’(except if the pain is enjoyable in some way).
