Absolutely.
To keep it at its freshest longest, make sure it's stored somewhere where it's not going to be exposed to lots of temperature changes (believe it or not, your bathroom is not a good place to store due to it getting steamed up when you shower, etc.). Also, take a look at the bottle - notice any discoloration? Then toss it - the fragrance has changed, or has faded.

Yes, perfume does lose its fragrance after awhile. That's why you should always be leery of buying it from those discount carts you see in the malls, they have old stock that may smell good when you first spritz it on, but 5 minutes later the fragrance is gone.

Actually, if you expose it to temperature changes, like say, having it in your bag in a hot car, perfume can lose its scent very quickly. Keep it away from windows and away from heat or cold to preserve it and it will last longer. If you've had it longer than 5 years, chances are it's time for a change.
