I dearly love Harlan Coben. He weaves a great story with a lot of suspense and twists and turns that keep you guessing right to the very last page. My favorite by him is Tell No One. Superb!!!
If you want a really, really light mystery, Janet Evanovich writes her Stephanie Plum series. It is always good for a laugh.
Charlaine Harris' Shakespeare series is great as well, if a bit dark in tone. OOH! And Sue Grafton is great as well!
AND!!! John Sandford's Prey series is top notch for thrills too.
AANNDD!! J. A. Konrath!! The Jack Daniels books are so so SO good! Kept me guessing...(!)
Shut me up now! There are just too many great authors out there to name only a few!

There are a lot of really good ones, who is best would be affected by the type of mysteries you like, historical, with psychological twists or more character or action driven. Some good ones are:
Anne Perry-historical
PD James-character driven
James Patterson-psychological twists
Sarah Paretsky
Tony Hillerman
Jonathan Kellerman
Arthur Conan Doyle---the Sherlock Holmes books
JD Robb--a cop in the future
Tess Gerritson
These are a few that I enjoy. Have fun reading.

The "best" of anything is a matter of personal taste. Try Edgar Allan Poe, Agatha Christie, Mickey Spillane, Ruth Rendell, Ellery Queen, Dashiell Hammett, Sir Arthur Conan Doyle, or Raymond Chandler. The Da Vinci Code might be categorized as a mystery and it has sold 65,000,000 books, one movie and numerous TV shows in 3 years. Agatha Christie has sold a billion books in english and a billion more in other languages, as well as the worlds longest running stage play.

There are many great mystery writers so it's hard to choose. Try these:
Patricia Highsmith: She wrote "Strangers on a Train" and the "Ripley series" of books. She can be dark and disturbing but you will become involved in the story and emotionally affected.
Ed McBain: Handles gory crime scenes with accurate prose and can be fall down funny. A great writer.

I see the classic ones have been mentioned, but if you want someone still alive, I think P.D. James is fabulous.
