You don't have to wait.
If you've already missed your period, just go a buy a test. Wait until morning and take it with the first pee you make. Your levels will be higher then and you will have the greatest chance of a good reading.
If it is negative, wait a week (if your period has still not arrived) and try again. By then you should definitely be able to get a true reading with a home pregnancy test. If it is still negative (and your period hasn't come) call up your regular doctor and schedule a pregnancy test. Make sure to tell the nurse that you've already taken the ones at home and the results were negative but, you still haven't had your period. They may opt for a blood test, which is definitive.

If you know when you could have conceived, if its been 1 week or 10 days before your period was expected then you can take the test now. If you don't know then take the test 1 week after you missed your period.

It mostly depends on the test you use, some tests say that you can take it right away, some say to wait til two days after. Read the box and see which one would suit you the best.
