A thesis statement tells what is the point of your essay or piece of writing. It summarizes the major claims you intend to make (and hopefully support) in one sentence or possibly two.

The key to an effective thesis statement is really knowing exactly what it is you intend to say or prove in your writing project. If you aren't sure of your conclusion it is impossible to distill it into one sentence.
