I think you would have to live in the region and attend conference games to answer this question honestly. I know Ohio State and Michigan is always a big game. I enjoy watching it each year.
However, I live in Louisiana and have been an LSU ticket holder since 1972. I have attended away games at all SEC schools. I have also lived in Birmingham, AL for two years and have many friends there. From that vantage point, I believe the Bama vs. Auburn matchup is a bigger game year end - year out.
My reason is that you have to understand that generally speaking the players and fans practically hate one another. In the State of Alabama you are either for Bama or Auburn and they talk about the game the entire year until next year's game.
Bama considers Auburn a bunch of rednecks and Auburn thinks of Bama as a bunch of stuck-up a**holes. Many fans buy a season ticket of the opposing school when their team plays away from home in order to get a ticket to the game. Now that's a pretty price to pay for one game.
I'm sure Ohio State and Michigan have their own stories to tell and I am sure the game is a heated riivalry but I am not so certain that it runs as deep as the Bama vs. Auburn game.

I think that it is definitely Auburn vs. Alabama. College football just has a different feel in the south. Michigan and Ohio State fans don't like each other, but they still respect each other. Auburn and Alabama fans HATE each other. If Auburn or Alabama win 1 game in a season, and that win is against their rival, that's a good season. Auburn-Alabama has also had some better moments, especially "Punt Bama, Punt" where Auburn blocked 2 punts late in the game for touchdowns to lead them to victory.

The entire state gets involved-people plan weddings around this game!
I went to a wedding once and the bride chose Iron Bowl weekend b/c of some reason, but realized that the game was on. So, she had TVs set up in the room the guys got dressed in (for the pregame shows) and had TVs set up through out the reception.
But, there were still groomsmen with radios in their ears for the service.
if she had not taken these measures, she may have not had anyone attend.

AND, the grooms cake at my sisters wedding had a Crimson Tide theme.

Michigan vs. Ohio State is always been a bigger game . both are big but this year this may decide the national championship.

the bigger game is the IRON BOWL. it aint just bigger, but year in and year out there've been more close games and dramatic victories than any other 2 schools can claim anywhere.
what make this game so big is the history between them and that both universities are in the same state - it's been played for a very long time, except during and after the war.
one could make a case for the ohio st/mich game being bigger this year ONLY b/c of their respective rankings.
otherwise and as you question states, year to year bama/auburn is as fierce as it gets.
