Usually I do it by saying, "Oh gross, there's sperm on your clothes!".

UV shows bodily fluids, Ninhydrin will detect protiens and their are kits which confirm semen via the acid phosphotase or Prostatic Specific Antigen present.

Use UV light. Body fluids fluoresce under UV light.
