from ShinOsaka station in Osaka to Tokyo station is 2 hrs 33 minutes to 2 hrs 37 minutes by the Nozomi Shinkansen according to Hyperdia.com If you click on "English" and enter ShinOsaka for start point and Tokyo in the destination it will show you this. It is about 2 hrs and 54 minutes on the Hikari Shinkansen. It will take 8 to 9 hrs on non-Shinkansen trains.
The flight from Osaka Airport to Haneda Airport in Tokyo is 1 hr 10 minutes.....plus checking in...getting luggage etc.
On the JR Highway bus
time of departure in Osaka 21:15 and 22:15
arrival in Tokyo 6:57 and 8:20.

As the person above says, its takes only two hours and thirty minutes by Nozomi Shinkansen. Air flight takes about an hour. You can take a flight either from Itami Airport or New Kansai Airport. You will land on Haneda Airport in Tokyo. From Haneda to downtown Tokyo, you can take a mono-rail train which takes about 20 minutes.

When I go home to the States for a visit i Fly on ANA (All nippon airlines) from Itami airport (Osaka) to Narita (Tokyo) then on to JFK (New york) the flight from Itami to Narita takes about 35 to 40 minutes. I don't know the cost as it is worked into my total flight plan. But the Plane is usually only about 30% full. You may want to look into this as an option.
