You gotta see Times Square at night! The Flickr link in the Sources field is a shot I took with my cell-phone camera last month; it's awe-inspiring in person. Lots of places to eat around Times Square, but the best reason is to go there is to see a show on Broadway. If you have ANY interest in theater, or just want great entertainment, you owe it to yourself to see SOMETHING.
I found myself scheduled on an unexpected trip to NYC on business a month ago, and because of connecting flight schedules coming home, I ended up with an extra night in Manhattan... so I splurged and bought a front-row ticket to "The Producers." It's hilarious -- well deserved all 12 Tonys it won -- and the St. James Theater is a classic institution in itself. If that's not to your taste, take a look, not only on the site I included, but search for other Broadway theaters and plays. I believe "Wicked" is back on stage (it was on a break while I was there, though I don't regret it given how much I enjoyed "The Producers"), and I also think "Spamalot" is still playing there.
What else? Just walk around town if you have time -- the park in Union Square is gorgeous, gawk at the Empire State Building (34th Street and 5th Avenue), go to the south end of the island and look at the Statue of Liberty, have a wood-fired pizza in Little Italy, you name it. That's the stuff I did when I wasn't working.
Is it expensive? YES. And you'll need cash for cabs (to get picked up, wave at the ones with the roof lights ON :-). The classic new-to-New-York experience is to look at the menu, see the prices, and think, "Now, how much is that in American money?" before you realize it IS in American money. And when I saw the room rate, I was REALLY glad work was picking up the tab.
But it's a fantastic city, filled with GREAT people from all over the world. Oh, and one thing y'oughta do -- it's something I didn't get a chance to do, and will make a point of doing if I go back: Walk up to an FDNY firefighter and shake his hand.

I would say, take a subway ride (above & below ground), take a taxi ride, walk around the south street seaport (money not required, but suggested), eat a REAL New York pizza (its not from any restaurant by the way), try your luck with a hot dog cart, see a broadway play (I suggest "Wicked"), walk through central park, oh, and one last thing - get lost in manhattan at least once!
