Can we say " 'In' a quiet and peaceful night"?
if it's wrong, what should I put in this blank? _____a quiet and peaceful night.
No matter how many times I prove read it I feel like something's wrong, but I'm not sure if it's right or not.

It depends if that is an ending or beginning of a sentence. As in:
"On a quiet and peaceful night, I walked in the woods."
or
"Nothing stirred in the quiet and peaceful night."
You would generally want to use "on" with the phrasing of "__ a quiet and peaceful night" or "in" with the "__ the quiet and peaceful night."
*Update*
After seeing your update, "On" is the only word that will correctly fit.

As one of the other answers notes, the first one is correct. However, if you want to ask a question, then the second one can be: "I'm not sure. Where can he work?" Hope that helps.

On a quiet, peaceful evening. Of course it does depend a little on what words precede and follow the phrase... Seeing your update, "On" is the correct word to use.

Look at it this way: Take out the adjs. "In a night." Does that make sense? Sounds better if it was "one" instead of "a," but if it needs the a, then try a verb.

"On". It sounds strange but if you insist on using 'quiet and peaceful night" the more attractive sentence would be " In the quiet of a peaceful night...".
"On a quiet and peaceful night..." sounds better grammatically in your context.
