Oh, there is so much there.
What do you like?
SHOPPING:
Lots of great shops. Fantastic prices. I can get loads of bargains in the street market near the clock tower. Clothes, gifts, crafts, foods, and decorative items galore.
HISTORY:
The castle, the cathedral, the astronomical clock, the Charles Bridge... so much to see! Do the Ghost Tour - it's great fun and is on every evening.
NIGHT LIFE:
Great clubs (there is a huge one near the Charles Bridge) and fantastic bars. Beer is the main drink here and is fantastic in price and quality.
FUN THINGS:
The waxworks is about £3 and well worth seeing. The puppet museum will have just re-opened. The river tours are supposed to be fantastic. I did a horse & cart tour which was fun. In have been told the zoo is good.
GREAT BUILDINGS:
Everywhere. Everything in the city centre is ancient and wonderful. Also lots of statues. TAKE A CAMERA YOU WILL NEED IT!
FOOD:
You can get any major food you want. I had a great pizza there. The Czech food is cheap and hearty. I am a vegetarian and had no problems at all. Restaurants are cheap enough to eat out every meal.
Any there is so much more.
I would stay in a hotel. Last time I went I stayed in a hostel and the place was very substandard (although it was £4 per night so I got what I paid for. LOL). I stayed in a 3* place first time I went and that was great. Where-ever you stay make sure you are within walking distance of the city centre, as although public transport is cheap and regular the system is not in English so getting off at the right stop may be tricky!

Yes, and lots do do / see. We were there in May coming from Russia and we stayed at the Pension "Veronika" south of town and at the end of a street-car line. It's about $ 50 or 60 a night, clean and a park across other side.

Whatever you do, don't stay in a hostel. They are really bad.
