It's hard to choose a best since she wrote so many great books, but several that are my favs are:
"And Then There Were None"
"The A. B. C. Murders"
"Murder on the Orient Express"
"The Murder of Roger Ackroyd"

I had a compendium of Hercule Poirot stories called Hercule Poirot's Casebook. It was fantastic.
You can get it on ebay right now for $7 including shipping. Item number 180003152851. This is not my auction. I just did a quick search.
EDIT
Ooh. Solstice has a point. The Murder of Roger Akroyd is a great one! If you want a full novel, go with that one. If you like short stories, do the Casebook.

'The Murder of Roger Ackroyd', with Poirot as the brilliant detective, is a good read - lots of twists, a devious mystery!
Also I agree 'And Then There Were None' (another name for Ten Little Indians) is good too.

Ten Little Indians -- they made it a movie too.
She was pretty much a formula writer, so you can't go wrong.
