Different conference affiliations really thats about it. Big 10 is primarily a Ohio Valley, or true midwest based confrence, while the Big 12 is centrally located and is not considered the midwest although most would say both conferences are formed by schools of the midwest. The Big 12 Conference is also a realtively new conference, although most consider it just an expansion of the Big 8.

All I know is that Big 10 universities are also called "Land Grant" Universities. This schools were founded by an act of congress in the 19th century. The purpose of the land grants was to finance this new schools, most of them in the Midwest. Never heard of Big 12.

One consists of schools mostly in the Great Lakes Region, while the other consists of teams in the Great Plains region. Only one state (Iowa) has schools in both conferences.
