As a fellow guitar player, you well know that, for the great ones, the guitar becomes an extension of the man and the ultimate outlet for the soul. Stevie Ray Vaughn was the poster child for this.
Jimi Hendrix
Eric Clapton
Eddie Van Halen
...are all excellent choices.
If you don't know about Stevie Ray Vaughn, check him out. Eric Clapton, once said, that SRV was the best guitar player! How about that.

Wow this question is popping up a lot. Once again i'll offer my answer.
The Best is Jimi Hendrix, because he could do stuff that nobody before or after him could ever do with a guitar. He had the most raw talent. Obviously some people don't like his music, but the music you play isn't what makes you the best, it's the way you play it. And no one was better than Hendrix at that.
Second is Duane Allman, for much the same reasons. an extremely innovative guitarist who didn't get to show us his true potential, but in the short time we were graced with his presence he earned himself the right to be up at the top of this list. Eric Clapton admitted he was better than him, so that's always a start.
Honorable mentions as always to the likes of Eric Clapton, Jimmy Page, Chet Atkins, Mark Knopfler, Stephen Stills, Slash, Carlos Santana, The Edge, Peter Green, and everyone else that other people will probably bring up.

Depends. Creatively, Jimi Hendrix. Versatility, Trey Anastasio. True representative of the rock n roll ethic and author of some of the best riffs ever to grace a record: Keith Richards. Fender soul with freedom of expression: Eric Clapton. Plus a huge number in between.

tony iommi of course he invented heavy metal in its self and has influenced years and years of guitarists and guitarists to come without him and his monstrous riffs music as we know it would sound very different.
Oh he played for black sabbath and has a solo band called iommi.
