If you want to go to both Sonoma & Napa Valley, I would stick to the southernmost part of both valleys, as there are proably 500+ wineries in the valley's combined. Ordinarily I would suggest two days, one for each valley. You can get there either from 101 over golden gate bridge if you want the scenic route. Or go right up highway 80 which is faster and better road.

Most people can handle 3 - 4 wineries in one day before either getting tired or being too drunk.

I would probably start at Domaine Carneros, in Napa, for a glass of sparkling wine and gorgeous view and lovely tour in the morning. Then I would go north up the napa side either highway 29 (heavy traffic) or silverado trail to either Silverado for some great sangiovese or sterling in Calistoga for great tram ride and views. Then I would probably pop over into the sonoma area in the late afternoon and check out Benziger, a cool bio-dynamic farmed winery that does some nice cab and chard.

That's a great loop through the valley and a bit of Sonoma.

Good luck!

Coming from SF, try to avoid Napa and the bottom of the valley. If you must go via Napa, try to take the Silverado Trail on the east side of town instead of the freeway on the westside.
You can approach upper Napa Valley either from the west via Novato to Sonoma or from the east via... (can't remember but I think it is) Green Valley which starts near the intersection of I-80 and I-680.
I like Sterling Winery and Mondavi, but I am not really qualified to suggest....
