Wow. I like how people get snarky with you, and then get the definitions wrong.

Sex is physical. Most simly, it's the parts you have. It gets more complex when you compare the physical things you can see with the chromosomes involved: there are 5 genetic sexes (not all of which I can remember, but combinations like XXY are possible along with the well known XX and XY).

Gender is more in the mind. People argue if there's biological factors influencing gender (transgender females share biological similarities in the brain with XX females). Others define gender as purely a social construction (it IS, definitely in large part, but I don't know if I would say that that's all it is).

Sexual preference is a misnomer for one. Sexual orientation is who you're attracted to. Male, female, anything that is neither / both, all of the above, NONE of the above....
It's generally seperate from both gender (and gender identity) and sex. It IS, however (not to be too contradictory), often considered a part of social gender to be heterosexual; as such, some argue that anyone who is GLB is non-gender-normative.

Sex either refers to sexual activity, or to being physically male or female (although some people are intersexed, meaning they have some of the physical characteristics of both males and females.)

Gender refers to being a man or a woman, which usually corresponds to being male or female. Some people develop an internal sense of gender that doesn't match their physical sex, though, and they try to live in the gender-role that matches their internal identity.

Sexual preference is simply which gender you prefer when it comes to sexual activity; whether you're excited by men or by women.

In a nutshell- (and I am not being flippant with this answer).

Sex is between the legs- whether you have a male or female body.

Gender is between the ears- whether you have a male or female mind and mentality.

Sexual orientation is who you want between the sheets- whether a male or a female.

A heterosexual male's sex is male, his mind is male and he usually desires persons of the opposite sex and gender.

A homosexual male's sex is male, his gender is male and he usually desires persons of the same sex and gender.

A male to female transsexual may have a male sex, but a female gender. Sexual orientation could be either heterosexual or homosexual. Sexual orientation is based on gender of the person in question and the gender they desire sexually, not on what sex the person in question is.
