Huh? The rooks first move of two squares? What are you talking about? Are you sure you don't mean the PAWNS first move of two squares? In which case the answer is no. The pawn can only capture on a forward diagonal move of one square. Or perhaps you meant castling in which case the answer is still no. Otherwise there is absolutely nothing special about the rooks first move, no matter how many spaces it is. So if it can reach the queen legally, of course it can capture it.
