I sincerely doubt that Klebsiella pneumoniae would grow in an ointment, but if it did, this could be determined by culturing the ointment on a suitable growth media in a clinical laboratory.

This would be little different from using a sputum sample. An agar plate would be inoculated and then observed. Any growth could be isolated and then identified.

Hmm...not sure if it could be present in ointment. Perhaps have a gram stain done on a sample of the ointment? Klebe is a gram negative organism, looks rather slender and has a capsule around it when one sees it gram stained.
