The chain in transfer cases drives the front wheels, so , no it does not turn in 2 wheel drive. To check for slack in the chain, put it in 4WD and lift both front wheels off the ground, and with the transmission in gear grab the front drive shaft and turn it. If it turns more than 1/16th of a turn, your chain is slack.
To find the clunk, have someone watch under your vehicle while you put it in low and hold the brake and let the clutch out a little. A bad mount will let the motor and transmission try to turn in the frame. Try it also in reverse.

purely to be sparkling, you've been having those noises OFF the line, good? The NV231 is area-time move case for off-highway use purely. used in 4 wheel drive on the line (or nicely-maintained airborne dirt and mud), the binding and crow hop that takes position may be interpreted by some people as clunking.

the chain disengages, the cluck is probobly because you "pop" the clutch. check your u-joints. that may be the source.
