Well, you don't say what kind of computer, what operating system, etc, etc... but I'll give you the answers on the assumption that you the commonest combination, a PC running Windows XP. The same principles apply to other variations too.
First thing to do is to run a thorough check for malware... viruses, trojans, spyware, which may be running in the background and using up resources. Use a reputable security tool/suite to do this.
Next, clean up your disk (s).... get rid of junk files, temporary files, browser caches, history files, old junk that you don't need. if you are using XP, there is a tool in "Programs/Accessories/System Tools" called "Disk Cleanup", which does a fair job of this.
In the same place, you'll find a disk defragmenter... when did you last run it?
Do you turn your machine off when you're not using it, or just leave it in standby? There are a number of good reasons for shutting it down at least overnight... not least being that you'll reset all the internal storage, and get rid of accumulating memory "leaks", which will slow you down.
Check your start files, see what's loading as soon as you boot... do you really need it all?
Consider adding more RAM - you can never have too much!

Scan it for viruses. Empty your Temporary Internet Files folder and any other temp folders if you can. Defragment your hard drive.

Run your defrag on your computer. Do a full scan on your antivirus. Or delete programs you don't use.

get a free copy of ccleaner. it works much better than windows disc cleaner to get rid of the excess junk. run the registry cleaner too. then get a free copy of spybot, adaware and a-squared. run them. what one might miss another will catch. then do a defragmentation of the hard drive.

try to restart it. if that doesnt work then try deleting the things on your computer that you dont use or need. if that still doesnt work then try to scan your computer.
