Have you written the Great American Novel? Expose? Biography? What's your book about? Have you had someone read it and critique it honestly? Do you know anyone who will give you a totally honest crititcism? Do you want to make a big splash in writing? Have you published before? How long is this book? Do you really want to have your work bought by a publisher or do you want them to represent it and pay you royalties as it goes into multiple printings?
That said, is your book any good? Does it tell a story that no one has read or heard before? Or, if it's a familiar one, have you told it with some new device that is irresistable?
Have you thought of self publishing? lulu.com is a source on the web. They'll publish anything and send you a finished product that looks professional... or as professional as you can make it when you send it to them in eMail or PDF file.

its a gonna take time, call publishing companies, convince them wid ur words & work and slowly its gonna work out, i suggest u start from writing stories for newspapers & magazines & then go for publishing companies coz they will be pleased if they see dat u r experienced in writing field, goodluck!!
