Oddly, mine are from two not so good movies and the line was the best part of the movie. I can't quote them exactly.
One is from a movie that had Jessica Lange, Diane Keaton and Sissy Spacek trying to figure their sister shot her husband and still trying to understand their mother's suicide with the family cat. They are constantly comparing the two incidences in terms of press coverage, etc. Sissy Spacek finally says something to the effect of why should I kill myself when I really just want "to put him out of my misery."
The second is form an old movie called Flight of the Navigator. A boy is kidnapped for study by aliens and they can't return him to the exact moment in time. They explain their tests and state obvious facts like humans don't use much of their brain, but when we tried to fill your brain up with data--we found it leaks!

"As you wish" from 'The Princess Bride'. Especially when the grandfather says it to the kid at the end- totally sealed it for me as a beautiful phrase. I love to say it to my loved ones who know the movie, when they're not expecting it.
Also, Fat B*stard in 'Austin Powers': "Get in my belly!!". You can't beat that.
I'll probably think of more.

I could sit here and type a whole list of great lines...I love movies.
probably one of my favorites, though is from the old classic, "It's a Wonderful Life"....because I think it's a line that rings true in reality...
"No man is a failure who has friends!"

I couldn't resist posting a quote from Star Trek and then Star Wars.
- I have been, and always shall be, your friend. Live long and prosper.
Spock's last words to Kirk.
“[Luke:] I can’t believe it. [Yoda:] That is why you fail.”

"Get with the beat, Baggy", from The Jungle Book.
I don't know why, it just makes me smile.
(I nearly said "You had me at hello" but someone beat me to it, and I had to do some hard thinking.)
Also: from Young Frankenstein, "I ain't got no body, and nobody cares for me, yatatata, yatatata, chah!" Makes me giggle every time.

(from When A Stranger Calls)
Oh--and "That is not my dog." from the Peter Sellers movie where someone asks him if his dog bites, and he says no, and then the dog viciously bites the guy, and the guy says "I thought you said your dog did not bite!" and he says "That is not my dog."
