The Colts will lose a game at some point this season. With parity (due to free-agency, salary cap, etc.), any team can win on a given week if it plays its best game - just look at the Bears vs. Dolphins this past week. The farther into the season the Colts go without a loss, the more motivated their opponents are going to be to be "the team" the knocks them off. Another factor is the distraction from the media as the season progresses. Some of the teams that have come close have had players say that the media attention was almost unbearable.
There is a reason that no team has accomplished this since 1972. In 1972, there was not a salary cap (so less parity) and the media scrutiny was only a fraction of what it is today.

No, simply because winning a Super Bowl is more important than an undefeated regular season. The Colts haven't been able to win one in a while despite them being so dominant in the regular season. They will be looking to win in this postseason and they aren't going to risk blowing their chances due to injuries playing games after they've clinched their division. I wouldn't be surprised if they took their superstars out not to risk it. That'll be the chance for another team to beat them.
