I would second The Simpsons. Though it depends on what you mean by "most" because The Simpsons has been running for a longer time than most of the other contenders but have a lot of episodes with no guest stars.
So perhaps a fairer comparison is guest star appearances averaged over episodes.
I'd still put my money on the Simpsons though.
