Barcelona (Spain) is definitely worth checking out! You can find a lot of things there: from art, architecture to nature...
Check out Parc Guell and La Sagrada Familia (Gaudi), museum of Miro, the Ramblas and especially enjoy the southern atmosphere.
Athens (Greece): the Akropolis, the museum of Cycladic art.
Gent (Belgium): many historical buildings, especially in the center, very nice to see.
I would suggest you to try limit the number of places you visit. Europe is rather big, and it's more worthwhile checking out a region instead of a quick jump from one big town to the next... You may see a lot of things these way, but miss the atmosphere of that country (or region)...
I went to Andalucia, in the southern of Spain during 3 weeks and even had the feeling I had to rush to see it all (which I didn't), and holiday is about enjoying too!

Hope you've got plenty of time.
I'd limit yourself to capital cities to fit in the really major sites.
If you flew into Heathrow and then got yourself a European train travel ticket, you could get round quite easily.
Starting in London (and depending on how long you have) you can go in a huge loop by going to Paris, Amsterdam, Berlin, Vienna, Prague, Athens, Rome, Madrid, then back to Paris and then London again.
Of these - not to be missed are London, Paris, Athens and Rome.

I'd suggest Spain, Italy, Greece, Malta (a small island) and all the other places in the Mediterranean sea, where it's warm (or shall I say hot) and very very nice :)

There are just many places to see, is this your first time? Well if it is then you have to go to Paris, it will just knock your socks off. It´s a really beautiful city, and if you are also traveling with your significant other it will be truely perfect.
Rome is also beautiful, and Barcelona is superb!
