The ONLY absolute specific test for semen is the presence of spermatazoa. I dried seminal stains the tails bteak away from the heads so what you look for is sperm heads. Usually a small portion of the suspect stain if teased apart in a drop of water and then dried usuall on a hot plate. This fixes the stain extract. The slide is then stained with haematoxylin and eosin for just a few seconds, rinsed and dried. A drop of mounting fluid is added to the slide and then a cover slip. When viewed under a good powerful microscope the sperm heads appear as charcateristic ovoid shapes with a distinctive acrosomal cap.

Stains are located on clothing, bedding etc usually by close visual examination. Although UV light is sometimes used to illuminate the material, this is rarely of any real use because of the widespread use of optical brighteners.

Often there is no visble staining and a chemical screening test is used. Filter paper is placed on top of different areas and dampened with distilled water. The paper is then removed and sprayed with a special chemical reagent. Seminal fluid contains very high concentrations of an enzyme called acid phospahatase. This catalyses the hydrolysis of one of the compnents of the reagent and a bright purple colouration is produced. Forensic laboartories make their own reagnets as this test is used frequently. Yoiu can buy kits for this purpose. They are readily available in the US. Usually they are marketed under the banner of 'supect your wife of infidelity? Other materials contain acid phosphatase but not to such a very high level. However, the test is only regarded as very strong indications of the presence of seminal fluid.

Immunological test are also available for the detection of a prostate specific antigen in dried stains.

CSI use an UV light. Works for most bodily fluids, even when it has been cleared up. Interesting Vom patterns found on a students floor.
