An operating system is the software which tells your hardware what to do. Hardware are the the things you can hit with a hammer (computer tower, monitor, etc, etc). Software is the thing you can only yell at, but not wack. The operating system is the thing that runs your programs and manages your hardware.
There are different operating systems. The most widely used and popular is Microsoft Windows®. Windows comes in different versions 3.1, 95, 97, 98, 2000, 2003, XP, Vista, etc. Another operating system is Linux which was built on Unix. Linux is a free as in freedom not free beer operating system. In other words you can do what you want with it, distribute it, copy it, reverse engineer it, throw out the window, flush it down the toilet bowl... Lots of fun stuff. And there is Mac OS, mac is the name of the operating system built on Unix (freebsd), plus mac looks cool and comes with names of cute fluffy animals like leopard, tiger, etc. Yeap, unix, linux are fun things.

Operating systems have a variety of native file systems. Linux has a greater range of native file systems, those being: ext2, ext3, ReiserFS, Reiser4, GFS, GFS2, OCFS, OCFS2, NILFS and Google File System. Linux also has full support for XFS and JFS, along with the FAT file systems, and NTFS. Windows on the other hand has limited file system support which only includes: FAT12, FAT16, FAT32, and NTFS. The NTFS file system is the most efficient and reliable of the four Windows systems. All the FAT systems are older than NTFS and have limitations on the partition and file size that can cause a variety of problems.

I'll keep it short...
See that tower or laptop sitting near you? Without an operating system its just a big expensive paper weight. The operating system tells your hardware to do stuff. Tells your software to run, tells your cd-drive to play music, tells your internet explorer window to browse to an internet site, and tells your sound card to play songs.

Operating system is a bult in program that runs in binaries such that we can use computer at higher lavel using known syntaces. If operating system is not present we have to write programs on binary format (i.e 0s and 1s)
