Oh no, what an impossible question as they are all so good.
If forced though, I'd have to chose the first one that I read - Equal Rites - because this is the one that got me hooked.
What do you think the best one is?

How can you choose just one? Terry Pratchett's books are all so unique and excellent. I just got the latest and am waiting for vacation in 2 weeks to sit down and become wrapped up in his world for 24 hours.

Good Omens (with Neil Gaiman). The two authors have very different styles but similar senses of humour; Gaiman's mythic reach gets rid of some of Pratchett's tweeness, while Pratchett's strong narrative drive offsets Gaiman's love of tangents. Also, it has a fantastic dog, and a protagonist called Anathema.

Personally, I've only read a few of his, but my favorite by far is Good Omens - he co-wrote it with Neil Gaiman, so technically I'm not sure if this is "a Terry Pratchett book", but it is my favorite.

My two favorites (sorry, I can't narrow it down to just one) are Witches Abroad, and Eric. Witches Abroad is one of the 'Ramtop Witches' books, on the whole I -really- enjoy that entire set of Pratchett's books but this particular one is my favorite out of all of them; Eric is a Rincewind book in which poor ol' Rince is mistaken for a demon of all things (and darn it, I just -love- Rincewind =) ... I've liked most Pratchett books I've read but these two in particular are "laugh out loud" books for me.

Pyramids is a much overlooked book in my opinion. Not weighed down with too much DiskWorld/Ankh Morpork baggage. A real story, with real ideas and it makes you think about what our own ancient Egyptians were like.

I have read them all.
Witches abroad is my favourite - typical brits abroad in a lot of ways.
I love most of his books, even the children's ones.

i would have to say nightwatch as it introduced me to such a fablious writer i have enjoyed all his books but not keenon the ones with ricewind in them
