John Steinbeck himself describes Lennie's condition as mental retardation. This condition can be described as gradational and so is multi-faceted. Mental retardation can be so mild as to be almost indiscernible - or so severe that the sufferer can hardly function.

Lennie's condition is severe in that he needs George to guide him and generally take care of him. His great bulk means that he can work and earn his keep - but Lennie would never be able to find that work for himself. He is therefore not self-sufficient. And whilst this gentle giant can suffer anguish at the fact that his overwhelming love has caused the death of a furry animal, he does not actually have a conscience. He has absolutely no concept that it is his strength which has caused the deaths - he doesn't learn from one death to another the consequences of the powerful pressure of his big hands.

However, this lack of conscience is through innocence. It does NOT, in Lennie's case, point to his having a psychotic nature.

His fixation on the "farm" might point in the direction of Asperger's syndrome - but since most sufferers of Aspergers also have a clever side to their condition (a propensity for art, music, maths, etc.), Lennie displays none of these attributes.

I have given three links. The first is to webmd's page of all mental health conditions, which you can research yourself for a match to Lennie's condition. The second is a fact sheet on mental retardation and symptoms - and the third is to the cliffsnotes page describing Lennie's character in full.

A good place to look for anything medical is WebMD. Don't use Wikipedia for anything that is scholarly, though.
