"40 foot contour interval " means that when you cross contour lines on a map, there is a 40 ft change in elevation. If you crossed 100 contour lines, you'd gain 40 * 100 = 4000 ft.

So the elevation gain will be found by multiplying the number contour lines crossed by the interval.
