There are a couple of explanations
His Wikipedia article states that Mrs Calabas was a reference to his late wife Jeanne Olson who died on Valentine's Day 1943. The couple lived in Calabasas, California for the last years of their marriage. Comedian Sonny King who knew the Durantes well said that Olson pronounced Calabasas as Calabash and it became an inside joke for the couple.
Another explanation that he was dining in a restaurant in Calabash, North Carolina in 1940 and liked it so much that he promised to make the owner Lucy Coleman famous. As he left, he told the owner "Goodnight, Mrs Calabash". The story claims that he started signing off his radio and television show with the catchphrase.
