open www.map24.de, enter as "start" Lehrter Strasse, Berlin and as "Ziel" (target) Museumsinsel, Berlin and then click on Route.

You can take a the bus line 147 from Berlin Hauptbahnhof to the bus station "Märkisches Museum/Inselstr." It takes about 15 min.
