Just say OK - that's your business - and then let her get on with her life.
Best mates can be a nuisance as they restrict your ability to experience a wider scene - and that's really important - get out and about and meet more people, then you can have lots of 'best mates'!

If she's your best mate then you should just carry on as before and give her the same hep/support/advice you gave her on her previous relationships.
You mention drugs though - I don't really understand that bit of your question. If she's got into some heavy drug scene then you need to talk to her about it and help her if you can.
You also mention depression - maybe you should advise her to see her GP if you think she's depressed.
But on the "how can I tell" part of the question - you can't. She may have discovered that she's a lesbian and this will be her lifestyle from now on. Or she may just be a bit mixed up (especially if drugs/depression are involved) and she will get a boyfriend next time round.
If you really care about her, just support her and do your best for her.
I hope she will find happiness, wherever it may be.
And I hope you do too:-)

Would you re-write your question so that those of us who only understand English can make any sense of it please? Something about lesbians drugs and depression but none of it strung together to make sense. Sorry. I'll have to pass on that one.
REALLY SORRY but as so many other people have obviously read AND understood your question so that they can give you an answer I can only assume that my system has failed and is missing out important parts of your question.

well when they fail each other, (like they've failed in man before) that would end, that should not necessarily be a lesbian relationship..They might as well be bi...Time would clear things.

Well, i think it is best just to try to encourage her about what you think is wrong about the relationship and try to get her to avoid the relationship.
