Joan Benoit Samuelson (born May 16, 1957) is an American former marathon runner who won gold at the 1984 Summer Olympics in Los Angeles. As a result she was the first ever women's Olympic marathon champion.:-)

Looks like you got the answer from others but I like this version better from the 1896 Olympics:
Less well known is the name of Stamata Revithi who can claim to be the first woman to have the run the course, just one day after Louis. Revithi was denied entry into the Olympic marathon because it was for men only but the following day she followed the same route, getting witnesses to verify her times at the start and at the finish which was where Evangelismos Hospital now stands.
It took her five and half hours but her achievement has never formally been recognised. There is also a story that a woman called Melpomeni ran four and half hours in one of the Olympics trials but was denied entry into the Olympic event.
