Yes you can! but this depends on you're PC motherboard and BIOS.

Some motherboards have a write-protect/read-only function which can be configured using the BIOS. However, common PC models have motherboards that do not support this.

You can't write protect a hard drive, only folders or files on the hard drive.

Why?

Every time you start your computer, your operating system has to access (and write to) your hard drive(s).

You can't protect an entire hard drive, or even separate partitions on a hard drive

The only way to do this is to turn your computer off and leave it off.

You might want to try creating a "Power-On" password in your BIOS.

Settings
Advanced
Prompt for password when computer goes off standby

you can't, really, and you have no reason to do so, it would render the disk more or less useless. you may be using the wrong terminology, what do you want to do?
