The mitotic index is defined as the number of mitotic figures (i.e. the number of cells in mitosis) per microscope field. In other words, if entry into mitosis is inhibited in some way, then the mitotic index will be very low compared to normal cells. On the other hand, if there is a mitotic arrest (because exit from mitosis is blocked) then the mitotic index will be very high relative to normal. Cells in mitosis can be identified with a variety of antibodies, We use an antibody specific for the phosphorylated form of Histone H3.

The mitotic index of the bone marrow of 7 healthy individuals was determined in Feulgen stained squash preparations to be 0.9% for the time 10 a.m. to 1 p.m. The method did not allow the distinction of mitoses of different cell lineageg. On the basis of this mitotic index, it is possible to estimate the average turnover time of the red cell series to be in the order of 11 to 33 hours and that of the white cell series to be in the order of 25 to 75 hours. In serial studies the mitotic index of 8 patients, accidentally exposed to various doses of ionizing radiation, was followed over a period of 4.5 months. A dose dependence of the mitotic index was noticed on the fourth post-irradiation day. After 4 weeks, the number of mitotic figures was within normal limits. After 4.5 morths the values were significantly higher than normal. (auth)

Mitosis is the process by which a cell separates its duplicated genome into two identical halves. It is generally followed immediately by cytokinesis which divides the cytoplasm and cell membrane. This results in two identical daughter cells with a roughly equal distribution of organelles and other cellular components. Mitosis and cytokinesis together is defined as the mitotic (M) phase of the cell cycle, the division of the mother cell into two daughter cells, each the genetic equivalent of the parent cell.
Mitosis occurs exclusively in eukaryotic cells. In multicellular organisms, the somatic cells undergo mitosis, while germ cells — cells destined to become sperm in males or ova in females — divide by a related process called meiosis. This occurs most notably among the fungi and slime moulds, but is found in various different groups. Even in animals, cytokinesis and mitosis may occur independently, for instance during certain stages of fruit fly embryonic development.
Transcription is generally believed to cease during mitosis, but epigenetic mechanisms such as bookmarking function during this stage of the cell cycle to ensure that the "memory" of which genes were active prior to entry into mitosis are transmitted to the daughter cells.
