If you say it with emphasis then it is something you do not want any part of sorta like "would you like some of my mystery meat stew?" and there are flies all over this rotten pot of meat and veggies or "would you like some malaria?"...that is a big NO THANKS.
But then say someone offers you a chocolate and you are on a diet...you really want it but you know it would derail all your efforts then you may say "thank you no" a bit more politely.

Actually you should say "No, thanks?"
The quotation marks should come affter the question mark, not before.
If you don't like my grammer lesson, just tell me "no, thanks."
