If I pinch myself with a needle that penetrates the third layer of the epidermis. Which is the layer that is going to get affected?

I think is the stratum basale layer or basement membrane.

I am not sure. Please help me. Thanks

Stratum Granulosum is the 3rd layer of skin from the surface:
The epidermis contains 5 layers. From bottom to top the layers are named stratum basale, stratum spinosum, stratum granulosum, stratum licidum, and stratum corneum.
So from top to bottom you have: Stratum Corneum, Stratum Licidum, Stratum Granulosum, Stratum Spinosum, Stratum Basale.

The third layer is stratum granulosum. The Stratum Basale is actually the bottom layer of the Epidermis.

The skin itself consists of the Epidermis, Dermis and Subcutaneous Tissue. All with different functions.
