Microsoft has an online scan at http://safety.live.com and TrendMicro has one called Housecall which used to be at http://housecall.trendmicro.com . Both will check for viruses and other malware.
After that you'll need to download some other protection to help keep it clean such as AVG (free.grisoft.com), ClamWin (clamwin.com), Ad-Aware (lavasoft.com), Windows Defender (microsoft.com)

AVG Free is my favorite virus scanner suite. It's done a better job than Norton on my computer. It's the first thing I install on a new computer.
For spyware, try out Spybot S&D:

AVG is junk. I recommend active virus shield. It is powered by kaspersky which is the number one rated antivirus. It is also free.
