If you want to know if someone else is a lesbian, ask her.
If you want to know if you are a lesbian then answer this quick checklist:
Do you want to f*ck women?
Do you have sexual feelings about women?
Do you ever touch yourself thinking of women?
If you answered yes to one or more of these questions then you are a lesbian or bisexual woman. If you answered no, then you are straight. Next ask yourself:
Do you want to f*ck men?
Do you have sexual feelings about men?
Do you ever touch yourself thinking of men?
If you answered Yes to one or more of these questions, then you are bisexual. If you answered no to all of the above then you are a lesbian.

It depends on who you think might be a lesbian. If it's you, then self-awareness will answer that question. If it's a friend, simply ask them.
