Well "first impression is the last impression" and in interviews the interviewer has less than an hour to judge your capabilities. So looks do matter but many will be of the opinion that it doesn't matter much. But you atleast need to be presentable i.e. well groomed. For e.g. visible facial hair and long hair for men is not favored in the corporate world. At the same time spectacles/glasses give the impression that the interviewee is a nerd (+ point in the corporate world.)
According to me "Dont judge a book by its cover," is hardly applicable at the time of interview since the time interviewer has to judge your talents and capabilities is very less.

Looks certainly matter in interviews.To be immaculately dressed is winning half the battle. But in the questioning session too you can't gaffe.But that is beside the point.From my experience,I can tell you that if you are sloppy,it will send the wrong signals to the potential employer.But don't overdo it to look too gaudy or garish.

Yes, look also matters in interview. Look does not mean you have to be like Shahrukh Khan or Aishwarya Rai.
It means:
1. a clean combed hair
2. a tucked in shirt
3. bright plain or neat checked shirts without any writings
4. Shoes or good pair of chappals [ no bathroom chappals pls ]
etc; etc; etc; etc;
