Something in a wave form such as light is passed thru or reflected off the materal being investigated. The difference in the amount of waves reflected or passed thru is compared to the amount of light originating from the source. A measurement is made, and usually put in a chart form.

What is measured is the difference between the amount of source light, and the amount of light that passed thru or reflected off the material being investigated.
