A one way trip Rio-Sao paulo will cost you US$35.00 and takes 6 hours.
Much safer to get a commuter flight, 35 minutes for US$85.00

It's about U$25. You can't book or pay it over the internet, but there are buses going from Rio to Sao Paulo every 20 minutes.
Just be aware it's not like the airports. People don't speak english there. If you really want do to this by yourself, write down the instructions in portuguese to get information.

Hi... fine answers, but perhaps you might want to consider a flight.. 40 minutes, and gets you from downtown Rio, to somewhat central Sao Paulo. These days you can get a very good deal on Varig Airlines.

About 400 miles you have to take the "Rodovia Dutra" nice ride but must be very careful specially if you are not familiar with the area, I travel to Sao Paolo every year, it's a beautiful city of 20 million people, lots of skyscrapers, oh and for that trip a car is better than the bus believe me..good trip..☺

Go to the bookstore right now and buy the Lonely Planet's Brazil book, if you haven't already. I'd say it's about 90% accurate when it comes to cost and time analysis of travel within Brazil.
But the prices range depending on what time of day you want to go. Sorry I dont know exact figures..

You can go to www.rodoviaria.com.br and look for sao paulo city.
you take the bus in the morning and get there in the night time... São Paulo look like New York.

Nonsense above. I make the trip every two weeks when I visit my girfriend.
Go with either 1001 or Itapemirim (I would recommend the latter, actually, because 1001 gets really busy). If you go 1001, make sure you buy Executivo or leito, lovely seats. If going by Itapemirim, buy a "golden" ticket.
Both of these are safe and very comfortable, tho at the bus station (rodoviaria), it pays to be vigilant. The journey takes about 6 hours. You can get to their websites by typing the company name and "onibus" into google. When buying your ticket, you can ask "Eu quero golden/executivo/leito para Sao Paulo. Que hora esta proximo onibus, por favor?"
If you have no time restrictions, I suggest travelling aroung 10 am - 12 noon as these times are less busy and you'll probably find you have two seats to yourself!

I would not recommend going by bus - unless you look like a local, can speak Portuguese fluently and are familiar with the trip. It will take you a couple days to make the trip. You're better-off going by rental car or motorbike. Beautiful country and very friendly people, but still there are bad apples in every bunch - be aware of your surroundings at all times (save the Caipirinhas for after the drive)
