Water pools in the low spot and it has started to leek.

It is a tar roof with a slight slant. The low spot is 1 or 2 feet from the drain.

This is why you have the low spot. The drain was not sealed properly and water has been getting in for years the the roof deck has rotted in that spot, that's why it is sinking. You need to rip all the roofing off that area and repair the deck first. Lots of fun.

The real trick is replacing the roofing so it will not leak again, and that depends on the type of roofing used. I made a built-up roof once which was still good as new after 25 years. Roll roofing is a slightly different story. You really need to know waht you are doing in something like this.

You could try, but it's a messy job. To do it correctly, you would strip off the low spot, build the spot back up with insulation, and rreapply the layers of roofing and tar.
As a quick fix, you can try using cold roofing tar and roofing felt. Cut pieces of the roofing felt to fit your low spot, gradually making your patch bigger with each layer. Then, start with a coat of the tar, then a layer of the felt, then tar, until you've built the low spot up. I'd top the final layer with some pea gravel just to keep the cold tar from making too much of a mess.
