An Amtrak train between the two cities is the fastest way to make the trip. It can take anywhere from 3.5 to 4 hours, depending on the schedule, and the fares also depend on time of day and day of the week. If you plan properly (Avoid taking the train on Friday or Sunday due to significantly higher fares.), you can make the round trip for under $150. Amtrak trains are pretty comfortable, so you will hopefully find that you can sleep on the train. That way, you can leave Boston early and return late, without worrying about lost sleep. Amtrak's Acela trains are not usually worth it. You pay a lot more than you do for Regional service, but you do not save very much time on such a short distance.
You can take a bus, and it will be cheaper, but it will also be slower and less comfortable. You should definitely consider it if you're on a budget, though. Flights are available, but you'd end up paying a lot, and you wouldn't save much time since you have to get to the airport in Boston so early, and the airports in New York City are far from all of the attractions, while Penn Station (where the trains arrive) is very close to many of them.

Yes, you can, but you need a full day, especially if you are relying on public transportation. Boston and NYC are about 214 miles apart. If you are driving or can get a ride, it's just about four hours, depending on how you go and whether there is heavy traffic or road construction. If you have to take public transportation, look for early morning buses and trains that will get you there before noon so you have plenty of time for sight-seeing. Watch the return schedule--be aware of the last bus or train back to Boston! Driving in cities like Boston and NYC can be a little scary if you are not an experienced driver. Parking can be expensive, too, but if you go with others you could split the cost. There are parking lots that will charge you around $25 or $30 to park for the full day. You may have a more restful trip if you take the bus or train, and the money you spend on the roundtrip ticket is offset by the price of gas, tolls, and parking.

I don't know why everyone is telling you to take a train. Amtrak is a ripoff. The commuter rails in each respective city are not so much of a ripoff (Boston's is the best for coverage and price I've seen in the USA). Either you fly out to NYC as was already suggested, or take a bus.
Greyhound is 65 or so I think for a single round trip between NYC and Boston, and there are companion fares. Also check out bonanza/peter pan bus lines online. One thing you might want to look into are Chinese buses between the cities. There are lots of odd bus lines that go between the two cities' Chinatowns. I live in Eastern CT and have taken a bus back to the local casinos for under 10 bucks, and got 20 dollars in coupons back to me upon arriving at the casino..
Just whatever you do, do not take the train for travelling between NYC and Boston.
If you're there for a while you might want to try taking the commuter rail from Boston to Providence, RI. It's a small city, roughly a third the size of Boston, with a lot of charm.
And if you're interested in going to one of the largest casinos in the world (in Connecticut), do not go to Foxwoods. Take it from a local: Foxwoods is poorly run, is a sloppy resort, and just isn't much fun compared to Mohegan Sun. Mohegan Sun is comparable to some of the best resorts in Vegas. www.mohegansun.com. If you do the casinos as a day trip from Boston, definately try to take a bus. They comp you out for more than what the round trip ticket costs.

The train is the fastest way to get from Boston to NYC and back, and that takes about 3 and a half hours roughly. Bus or car would take much longer, but if you took a train early in the morning, you could easily be there late morning/mid day, spend the entire day in NYC and then take the train back. Just make sure to buy your tickets ahead of time, and leave enough time to get to the train station, etc. People do it a lot, so I don't think you'd have a problem.

Mass neighbors NY so the drive is 3-4 hours but don't just stop there. There are many day trips to Cape Cod, Mystic Seaport, Hampton NH, Mohegan Sun, Foxwoods Casino, Suffolk Downs. You will be centrally located in NE so enjoy all that you have time for.

Personally I like Providence, which is in Rhode Island but still sort of between Boston and NYC (if you drive up I-95 instead if I-84). They have good pizza there so you can get plenty of slices. Unless you're referring to something else, in which case, you can get that too. Between the clubs, bars, restaurants, and universities there, there's a lot going on in Providence for a city its size. And plenty of females. I also forgot the casinos in Connecticut - Foxwoods in particular.

Flights on Jet Blue run about $50 each way + taxes, so it may be cheaper than Amtrak. There's a Chinatown bus that runs between Boston's Chinatown & NYC's Chinatown that is under $20 each way. Don't know the name, but I have friends who have used it.
