It is a name for a type of defense.
Teams usually run the 4-3, that is, 4 down (with the hand on the ground prior to the snap of the ball) linemen, and 3 linebackers (the three guys who stand about a yard or more off the line of scrimmage(where the football is before the snap).
The other 4 are Defensive Backs, either safeties or cornerbacks, usually 2 of each.
In the last few years, some teams have started to run the 3-4, like the Jets.
So, the 3-4 just refers to (in order) the number of down linemen and the number of linebackers (standers).

The 3-4 is the number of lineman and line backers. Something special about the 3-4 is that because of the minimum number of lineman, they tend to be bigger in size, as seen in the chargers and cowboys. The nose tackle (center lineman) is often the biggest.
In addition, there's the 4-3 which shows 4 lineman 3 linebacks. The nickel which is 4 lineman 2 linebackers and 5 defensive backs. The dime which is only 1 linebacker and 6 defensive backs, and the quarter which is 3 lineman, 1 linebacker and seven defensive backs.

It is when there is 3 linemen on the line of scrimmage on the defensive side of the ball and 4 linebackers behind them. Those 7 plus the deep 4 safeties make up the 11 man defensive squad

the 3-4 is a style defense played in football.you can only have 11 men on the feild so a 3-4 implies 3 lineman, the big guys who start with there hand on the ground, and 4 linebackers then you have 4 db the guys who stop the other team from catching the ball.It is usually played when a team has numerous good linebackers or good d lineman or on plays were you know the other team will throw so you have more guys covering recivers.

There are three defensive linemen, four linebackers, two safeties (one strong, and one free), and two corners in the 4-3 defensive set.
