I think you know the answer to this. Even if you were not married, it would not be a good idea to get involved with your daughter's 21 year old friend. The fact that you are married, makes her behavior even more inappropriate. It is time to be the adult here and tell the girl to quit it!

Oh that's got to be awkward. I would talk nonstop about your wife and refer to your daughter a lot. The more she realizes you're a husband and a father the less she may be interested.

I would have to put her in her place, tell her you do not appreciate her demeanor and that she should have more respect for her elders. I would also go as far to tell her that she makes me feel uncomfortable and that she makes you feel like you do not want to come visit your daughter.

Ask your wife to come with you and ask your wife to stick up for you. My wife had a friend hitting on her through email, and she asked him to stop, but then one day I emailed him saying he needs to treat her with respect. He has refused to even speak to her since. Usually when someone is doing that, they don't realize they're being disrespectful, and when the person who they are flirting with asks them to stop it can sound playful, but when they bring in the other person, the other person saying "Back off," really makes an impression.
