The subject of the sentence is "you."
As in (re-ordering the words):
You (subject) do spell (verb) that (object) how (adverb, modifying verb)?
Compare:
I (subject) do make (verb) that (object) quickly (adverb, modifying verb).
Or
Quickly do I make that.

i think your subject in the sentence "can someone please tell me where the subject of this sentence is?" i believe is the question itself. 'how do you spell that' i would say the person you are talking about because you are saying how do they spell it, not how is it spelled.

The sentence form of this would be: You do spell that, so:
You= subject
