Edward V's brief reign came to an end on 26th June, when Parliament accepted Gloucester's claim that Edward IV's marriage was invalid and his children illegitimate. It proclaimed Gloucester King Richard III and, soon afterwards, the two princes disappeared from the Tower. It is possible they were murdered by Richard's agents in August 1483, but responsibility for the crime has also been attributed to Henry Stafford, Duke of Buckingham, and to Richard's successor, King Henry VII. Skeletons found in the Tower in 1674 are thought to be those of Edward and his brother.

Not Richard III, but someone who thought he would be doing him a favor. From what I understand, Richard III was a good man, and wouldn't have wanted to cause anyone's death, esp. his nephews.

There weren't too many people who has access to them in the tower. There bodies were autopsied and they were believed to have been starved to death. Therefore it had to be Richard III. He was the only one that benefited from their death.

We are not certain they were killed. The bones found in the tower were buried 10 feet deep. This is the Anglo/Roman level. Probably Saxon foundation sacrifices. Pagans did that. The elder boy was being treated for a jaw infection. Why would Uncle Richard allow medical attention if he wanted him dead? The jaw infection may have killed him at a later time. I think The Duke of Buckingham did it. He was notorious for wanting be King himself.
