If you go to Toonopedia, you will be able to verify the answers that people give you on this topic. I remember reading about Obadiah Oldbuck, and there was also something in England called Alley Sloper, though that may have been a newspaper strip.

I don't know about the first comic BOOK, but the first superhero comic STRIP was The Phantom, by Lee Falk in the 1930's. There were some other comic strips before this, but they didn't involve superheros.
